RESUMO

BACKGROUND: Chronic bovine mastitis is linked to biofilm-producing Staphylococcus aureus (bp-Sa) or Staphylococcus coagulase-negative (bp-Scn). OBJECTIVES: Bp-Sa and bp-Scn were treated with intramammary preparations of either enrofloxacin HCl·2H2O-dimethyl-sulfoxide-chitosan (enro-C/DMSO/chitosan) or enro-C alone. Their potential to inhibit and degrade biofilm formation in vitro was also assessed. METHODS: Milk samples were obtained from the affected quarters in a herd. Phenotypical and genotypical identifications as biofilm-producing Staphylococcus species were carried out. Enro-C/DMSO/chitosan and enro-C alone were assessed to determine their in vitro efficacy in interfering with biofilm formation and their bactericidal effects. A prolonged eight-day treatment with a twice-daily intramammary insertion of 10 mL of enro-C/DMSO/chitosan or enro-C alone was set to evaluate the clinical and bacteriological cures on day 10 in 15 cows per group and the biofilm-inhibiting ability. RESULTS: Fifty-seven percent of the isolates were identified as Staphylococcus spp., of which 50% were bp-Sa, 46% bp-Scn, and 4% Staphylococcus pseudintermedius. One hundred percent of the S. aureus isolated and 77% of Staphylococcus coagulase-negative were biofilm producers. In both groups, the icaA and icaD biofilm-producing genes were identified. The experimental preparation could inhibit biofilm formation, degrade mature biofilms, and have well-defined microbicidal effects on planktonic and biofilm bacteria. The respective clinical and bacteriological cure rates were 100% and 80% for enro-C/DMSO/chitosan and 41.7% and 25% for enro-C alone. CONCLUSIONS: Enro-C/DMSO/chitosan eliminates bp-Sa and bp-Scn from cases of chronic bovine mastitis.

RESUMO

Natural populations of the spider crab Maja brachydactyla constitute a fishery resource of great economic importance in many countries. As in the rest of eubrachyurans, the females of this species have ventral-type seminal receptacles where they store sperm from copulations. Sperm can be stored in these structures for months and even years before egg fertilisation, with the consequent degradation of the sperm cells during the time. In this work, we analyse the viability and the possible genetic damage in sperm accumulated in the seminal receptacles of M. brachydactyla females as a function of the storage time (from 0 to 14 months) using the comet assay technique. On one hand, we developed an algorithm for comet image analysis that improves the comet segmentation compared with the free software Open comet v1.3.1 (97% vs. 76% of detection). In addition, our software allows the manual modification of the contours wrongly delimited via the automatic tool. On the other hand, our data show a sharp decline in sperm viability and DNA integrity in the first four months of storage, which could lead to a decrease in the fecundity rate and/or viability of the embryos or larvae from the second and third clutches of the annual cycle if the repair capacity in these gametic cells is low.

RESUMO

The present study examined factors that could be associated with bias victimization according to intergroup threat theory, namely socioeconomic status (SES), acculturation (Anglo orientation and Latino orientation), immigrant status, and their interactions. Self-identified Latino participants (N = 910) from three cities in the United States were queried about experiences with bias victimization, specifically hate crime and noncriminal bias victimization. Findings revealed that levels of bias victimization, hate crime, and noncriminal bias victimization were associated with SES, Anglo orientation, immigrant status, and their interactions, although in some unpredicted ways. Interactions among key variables helped clarify the roles of these factors in concert on bias victimization. The hate crimes against U.S.-born Latinos and the victimization risk associated with increasing Anglo orientations among immigrants contradicts predictions of intergroup threat theory. More nuanced analyses of social locations are needed to examine bias victimization.

RESUMO

INTRODUCTION: The Latin American Society of Pediatric Infectious Diseases (SLIPE by its Spanish acronyms) is working to understand the current situation, gaps, and opportunities for traceability of the quality vaccination process in Latin America and the Caribbean. AREAS COVERED: On September 24th and 25th, a Latin American forum of experts in immunization programs was held through the Zoom platform; the topics discussed included: computerized systems for recording immunizations, vaccination programs traceability, challenges, and information systems for the integrated management of vaccination. EXPERT OPINION: Latin American countries have transitioned from having a nominal registration system to a nominal tracking system, with many of them not migrating their platforms to new technologies; therefore, the low-quality data, fragmented databases, and slow information traffic present a challenge that must be taken on.

RESUMO

We studied the trends of height-for-age (HAZ) Z scores by socioeconomic status (SES) groups of Chilean boys and girls aged 5-18 born between 1877 and 2001, by performing a meta-analysis of 53 studies reporting height-for-age sample data from which 1258 HAZ score datapoints were calculated using the 2000 reference growth charts for the US of the Centers for Disease Control and Prevention (CDC). We found stagnant mean and median HAZ scores of about -1.55 to -1.75 for the general population, and -2.2 to -2.55 for lower SES groups up to cohorts born in the 1940s. However, we found an upwards structural change in cohorts born after the 1940s, a period in which HAZ scores grew at a pace of about 0.25 to 0.30 HAZ per decade. Since this change happened in a context of moderate Gross Domestic Product (GDP) growth, high and persistent income inequality, and stagnant wages of the working class, we discuss the extent to which our findings are associated with the increase in public social spending and the implementation and expansion of a variety of social policies since the 1940s and early 1950s.

RESUMO

BACKGROUND: The risk of severe coronavirus disease 2019 (COVID-19) varies significantly among persons of similar age and is higher in males. Age-independent, sex-biased differences in susceptibility to severe COVID-19 may be ascribable to deficits in a sexually dimorphic protective attribute that we termed immunologic resilience (IR). OBJECTIVE: We sought to examine whether deficits in IR that antedate or are induced by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection independently predict COVID-19 mortality. METHODS: IR levels were quantified with 2 novel metrics: immune health grades (IHG-I [best] to IHG-IV) to gauge CD8+ and CD4+ T-cell count equilibrium, and blood gene expression signatures. IR metrics were examined in a prospective COVID-19 cohort (n = 522); primary outcome was 30-day mortality. Associations of IR metrics with outcomes in non-COVID-19 cohorts (n = 13,461) provided the framework for linking pre-COVID-19 IR status to IR during COVID-19, as well as to COVID-19 outcomes. RESULTS: IHG-I, tracking high-grade equilibrium between CD8+ and CD4+ T-cell counts, was the most common grade (73%) among healthy adults, particularly in females. SARS-CoV-2 infection was associated with underrepresentation of IHG-I (21%) versus overrepresentation (77%) of IHG-II or IHG-IV, especially in males versus females (P < .01). Presentation with IHG-I was associated with 88% lower mortality, after controlling for age and sex; reduced risk of hospitalization and respiratory failure; lower plasma IL-6 levels; rapid clearance of nasopharyngeal SARS-CoV-2 burden; and gene expression signatures correlating with survival that signify immunocompetence and controlled inflammation. In non-COVID-19 cohorts, IR-preserving metrics were associated with resistance to progressive influenza or HIV infection, as well as lower 9-year mortality in the Framingham Heart Study, especially in females. CONCLUSIONS: Preservation of immunocompetence with controlled inflammation during antigenic challenges is a hallmark of IR and associates with longevity and AIDS resistance. Independent of age, a male-biased proclivity to degrade IR before and/or during SARS-CoV-2 infection predisposes to severe COVID-19.

RESUMO

Class III plant peroxidases (Prxs) are involved in the oxidative polymerization of lignins. Zinnia elegans Jacq. Basic peroxidase (ZePrx) has been previously characterized as capable of catalyzing this reaction in vitro and the role in lignin biosynthesis of several of its Arabidopsis thaliana homologous has been previously confirmed. In the present work, ZePrx was overexpressed in Nicotiana tabacum to further characterize its function in planta with particular attention to its involvement in lignin biosynthesis. Since Prxs are known to alter ROS levels by using them as electron acceptor or producing them in their catalytic activity, the impact of this overexpression in redox homeostasis was studied by analyzing the metabolites and enzymes of the ascorbate-glutathione cycle. In relation to the modification induced by ZePrx overexpression in lignin composition and cellular metabolism, the carbohydrate composition of the cell wall as well as overall gene expression through RNA-Seq were analyzed. The obtained results indicate that the overexpression of ZePrx caused an increase in syringyl lignin in cell wall stems, suggesting that ZePrx is relevant for the oxidation of sinapyl alcohol during lignin biosynthesis, coherently with its S-peroxidase nature. The increase in the glucose content of the cell wall and the reduction of the expression of several genes involved in secondary cell wall biosynthesis suggests the occurrence of a possible compensatory response to maintain cell wall properties. The perturbation of cellular redox homeostasis occurring as a consequence of ZePrx overexpression was kept under control by an increase in APX activity and a reduction in ascorbate redox state. In conclusion, our results confirm the role of ZePrx in lignin biosynthesis and highlight that its activity alters cellular pathways putatively aimed at maintaining redox homeostasis.

RESUMO

Comparison of emotion perception in music and prosody has the potential to contribute to an understanding of their speculated shared evolutionary origin. Previous research suggests shared sensitivity to and processing of music and speech, but less is known about how emotion perception in the auditory domain might be influenced by individual differences. Personality, emotional intelligence, gender, musical training and age exert some influence on discrete, summative judgments of perceived emotion in music and speech stimuli. However, music and speech are temporal phenomena, and little is known about whether individual differences influence moment-by-moment perception of emotion in these domains. A behavioral study collected two main types of data: continuous ratings of perceived emotion while listening to extracts of music and speech, using a computer interface which modeled emotion on two dimensions (arousal and valence), and demographic information including measures of personality (TIPI) and emotional intelligence (TEIQue-SF). Functional analysis of variance on the time series data revealed a small number of statistically significant differences associated with Emotional Stability, Agreeableness, musical training and age. The results indicate that individual differences exert limited influence on continuous judgments of dynamic, naturalistic expressions. We suggest that this reflects a reliance on acoustic cues to emotion in moment-by-moment judgments of perceived emotions and is further evidence of the shared sensitivity to and processing of music and speech.

RESUMO

Inulin-type fructans are polymers of fructose molecules and are known for their capacity to enhance absorption of calcium and magnesium, to modulate gut microbiota and energy metabolism, and to improve glycemia. We evaluated and compared the effects of Chicory inulin "Synergy 1®" and inulin from Mexican agave "Metlin®" in two experimental models of colon cancer and bone calcium metabolism in mice and rats. Inulins inhibited the development of dextran sulfate sodium-induced colitis and colon cancer in mice; these fructans reduced the concentration of tumor necrosis factor alpha and prevented the formation of intestinal polyps, villous atrophy, and lymphoid hyperplasia. On the other hand, inulin treatments significantly increased bone densitometry (femur and vertebra) in ovariectomized rats without altering the concentration of many serum biochemical parameters and urinary parameters. Histopathology results were compared between different experimental groups. There were no apparent histological changes in rats treated with inulins and a mixture of inulins-isoflavones. Our results showed that inulin-type fructans have health-promoting properties related to enhanced calcium absorption, potential anticancer properties, and anti-inflammatory effects. The use of inulin as a prebiotic can improve health and prevent development of chronic diseases such as cancer and osteoporosis.

RESUMO

Abstract: Background: This systematic review aims to report the current knowledge of retinoblastoma (Rb) and its implications in Mexico. We analyzed clinical and demographic data of patients with Rb at select hospitals with Rb programs or that treat and refer patients with Rb, and identified the gaps in practice. We propose solutions to improve diagnosis, provide adequate treatment, and improve patient uptake. Methods: A general review was conducted on PubMed of peer-reviewed literature on Rb in Mexico. Ophthalmology Department Heads or Directors of Rb programs at seven hospitals in Mexico were contacted for data available on their patients with Rb. Results: Five hospitals provided clinical data on 777 patients with Rb in a period spanning 2000-2015. Of the 122 patients with treatment, 83.4% underwent enucleation. From 33 to 45.3% of Rb tumors in Mexico reach an advanced intraocular stage of development. Knowledge of the disease is limited, despite the fact that the Mexican Retinoblastoma Group has elaborated Rb treatment guidelines and is developing a national Rb registry. Especially in the Southern states, prevalence and outcomes are comparable to African and Asian countries, and only few patients are referred to national treatment centers. Only three institutions have comprehensive Rb programs. Conclusions: There is an immediate need in Mexico to expand primary care providers' knowledge of Rb and to expand and upgrade current Rb programs to meet the needs of the population adequately. Diagnosis and care of Rb patients in Mexico can also be improved by the establishment of a national Rb registry and a national early detection program, and by increased use of the national treatment protocol.

Comparing the slopes of aqueous-based and standard addition calibration procedures is almost a daily task in analytical laboratories. As usual protocols imply very few standards, sound statistical inference and conclusions are hard to obtain for current classical tests (e.g., the t-test), which may greatly affect decision-making. Thus, there is a need for robust statistics that are not distorted by small samples of experimental values obtained from analytical studies. Several promising alternatives based on bootstrapping are studied in this paper under the typical constraints common in laboratory work. The impact of number of standards, homoscedasticity or heteroscedasticity, three variance patterns, and three error distributions on least-squares fits were considered (in total, 144 simulation scenarios). The Student's t-test is the most valuable procedure when the normality assumption is true and homoscedasticity is present, although it can be highly affected by outliers. A wild bootstrap method leads to average rejection percentages that are closer to the nominal level in almost every situation, and it is recommended for laboratories working with a small number of standards. Finally, it was seen that the Theil-Sen percentile bootstrap statistic is very robust but its rejection percentages depart from the nominal ones (<5%), so its use is not recommended when the number of standards is very small. Finally, a tutorial and free software are given to encourage analytical laboratories to apply bootstrap principles to compare the slopes of two calibration lines.

RESUMO

This paper studies the trends in height-by-age across socioeconomic groups of Chilean boys aged 5-18 born between 1880 and 1997, by performing a meta-analysis of 38 studies reporting height-by-age published since 1898. We estimate the trends using quantile regressions and by analyzing detailed height data from five selected studies. Both methods yield an average decennial increase in height of 1-1.1cm, and 0.9 and 1.2-1.3 cm for boys of upper and lower socioeconomic status (SES), respectively. SES differences in heights of 9-11 cm are observed up to the late 1940s. However, boys born after the 1930s exhibit substantial convergence in height between socioeconomic groups, driven by an increase in height of middle and lower SES boys of 1.5 and 1.4-2 cm per decade, respectively. As a result, SES differences in height decreased to 5 cm in 1990s. Since these changes occurred in a context of moderate economic growth and persistent income inequality, we argue that our findings are associated with the emergence and expansion of social policies in Chile since the 1940s, which delivered steady improvements in health, nutrition and living conditions.

Abstract: Arch Med Interna 2013 - 35(1):05-08 Neurological complications are common in patients with human immunodeficiency virus (HIV). We conducted a descriptive study, to determine the frequency of neurological disease in HIV/AIDS (acquired immunodeficiency syndrome) in our center, their clinical and developmental profile, and compare the degree of immunosuppression, the length of hospital stay, severity of the condition (estimated income requirement to intensive care unit [ICU]) and mortality between the patients presenting with opportunistic disease (OD) and non-opportunistic disease (NOD). High prevalence of neurological disease (21%) was found, mostly in young people, male, with severe immunosuppression, dominating the AIDS stage. ODs are more frequent and have higher recorded morbidity than NOD. Our population does not record the epidemiological transition seen in the developed world linked to the use of antiretroviral therapy (ART), a fact we attribute to the socio-economic-cultural situation.

RESUMO

The potential wound healing ability of sodium alginate (SA) meshes was determined in cavitated rat wounds infected either with Escherichia coli or Staphylococcus epidermidis. Wound progress was evaluated macroscopically and histopathologically to assess dissemination of the induced infection through evaluation of granuloma formation and adjacent tissue irritation. Five groups of 24 female Wistar rats weighing 280 ± 34 g were formed as follows: control group with SA mesh without inoculum, control group with Escherichia coli inoculum without SA mesh, control group with Staphylococcus epidermidis inoculum without mesh, group with SA mesh + Escherichia coli inoculum and group with SA mesh + Staphylococcus epidermidis inoculum. Abdominal flank incisions were performed under anaesthesia and groups were formed. Both macroscopic and histopathological follow up were carried out and results indicated that the presence of SA meshes avoids or greatly diminishes bacteria dissemination and adjacent tissue irritation. This reaction occurs even in the presence of Escherichia coli or Staphylococcus epidermidis. In the control groups with SA mesh without inoculum and in the groups with Escherichia coli inoculum without SA mesh, and control group with Staphylococcus epidermis inoculum without mesh and group with SA mesh + Escherichia coli inoculum re-epithelialization and wound healing aspect were better than in groups inoculated with Staphylococcus epidermidis, the general odd ratios showed that the dissemination risk was 58 times greater in treatment with SA mesh and E. coli as infectious agent (IC 95% 6.2-210). These results indicate that SA mesh is capable of providing adequate conditions to allow granulation of cavitated wounds, without irritation to adjacent tissues and avoiding dissemination of the infection by E. coli and Staphylococcus epidermidis in rats. However, presence of granuloma can be a healing or esthetic disadvantage, as observed in control group with SA mesh without inoculum.

The introduction of exogenous genes into plants permits the development of a new generation of biological products, i.e., edible vaccines. Cereals, especially maize, have been the systems of choice for the expression of antigenic proteins because the proteins can be expressed at high levels in the kernel and stored for prolonged periods without excessive deterioration. The utilization of plant-derived antigens for oral delivery provides an alternative strategy for the control of pathogens in animals compared to the current vaccine administration methods, such as injection. However, there is some doubt about the efficacy of these types of vaccines in polygastric animals due to the features of their digestive system. Here, we report the efficacy of an edible vaccine against rabies evaluated in sheep. Kernels containing different doses of G protein (0.5, 1, 1.5 and 2mg) were given in a single dose by the oral route. Cumulative survival was better in groups that received 2mg of G protein and for the positive control (inactivated rabies vaccine); this observation was supported by the presence of neutralizing antibodies. Animals in the control group died after challenge. The degree of protection achieved for 2mg of G protein was comparable to that conferred by a commercial vaccine. In conclusion, this is the first study in which an orally administered edible vaccine showed efficacy in a polygastric model.

RESUMO

Animal production systems in pursuit of sustainability production, have to integrate several elements such as: environmental protection, food safety, animal welfare, and benefits to local producers. The objectives of the study were: 1) identify the attitudes of consumers in three districts of Mexico City towards sustainable attributes; 2) identify their perceptions regarding the production of animal food products; 3) determine if there is an effect of the social-demographic variables. In this sense, one thousand two hundred consumers were surveyed in markets of three districts of Mexico City with different social-economic levels. Results show that attitudes and perceptions of consumers differ mainly according to the district where they reside. In relation to the sustainable attributes, consumers considered food safety as the most important attribute of an animal product, followed by environmental protection. Regarding animal food production in Mexico, consumers perceived in first place that this benefits local producers, that they are safe products, that it improves animal welfare, and, finally, that it conserves the environment. These results revealed that consumers in Mexico City show positive attitudes towards sustainable animal food production attributes, which could represent new opportunities for the food industry in Mexico.
